POSITION SUMMARY:

The Research Technician will be a key contributor to the development and execution of innovative anti-aging intervention projects. The Technician performs husbandry, housing and care of rodents within the vivarium.

Duties and responsibilities:

  • Set up housing and provide feed and water.
  • Monitor and ensure appropriate photoperiod cycle daily: setting and assessing room mechanical timer for appropriate operation on 24-hour cycle for 14-hour day and 10-hour dark cycle.
  • Track and document temperature and humidity ranges every 24 hours within the holding room with attention to the Guide for the Care and Use of Laboratory Animals standards for acceptable ranges.
  • Operate within a noise and vibration monitored environment to protect research models from disruptive confounding and breeding sensitive impact.
  • Capable of observing animals for general health and general animal room standards.
  • Recognize deviations from normal or expected outcomes based on known reference standards and makes recommendations to supervisor.
  • Recognize and document unusual occurrences and notify supervisor.
  • Report all animal care concerns and become involved in the resolution and appropriate corrective action when necessary.

The Opportunity:

Mitochondria are dynamic organelles vital to several functions within the cell. Decline in mitochondrial function is a distinctive feature in several age-related pathologies including sarcopenia, metabolic dysfunctions, and neurodegenerative disorders. It has been observed that, with age, mutations in the mitochondrial DNA accumulate at an order of magnitude higher compared to the nucleus. The Boominathan lab at SENS Research Foundation is developing gene therapies to functionally express the 13 protein coding genes in the mitochondrial genome from the nucleus (also called allotopic expression). We are also interested in modulating the mitochondrial DNA for sustained organelle fitness and function over time. Using disease cell lines and in vivo models, our lab integrates molecular biology, biochemistry, and computational tools in achieving these goals. Key Responsibilities:

This research position is within a collaborative and dynamic group developing transitional therapies for mitochondrial dysfunctions.

  • Plan, design and execute all aspects of preclinical research

Technical Skills and Qualifications:

We seek a postdoctoral fellow to join our small but dynamic immunology team led by Dr. Amit Sharma for a project geared toward investigating the mechanisms involved in the age-dependent decline in immune surveillance of senescent cells with the aim of finding promising interventions.

There are three main projects currently for the postdoctoral fellow. One of the projects involves characterizing age-dependent phenotypes changes in the of Natural Killer cells and its implication on their ability to eliminate senescent cells in cell culture and mice model. We have identified several unique antigens expressed on the surface of senescent cells. The goal of the second project is to characterize the surface antigens on senescent cells and with the goal of developing CAR-NK cells with therapeutic application. The projects will require validating the CAR-NK cells in cell culture and the mice models of senescence. Senescent cells are known to secrete a unique mixture of proinflammatory cytokines, chemokines and matrix modifying proteins called SASP (Senescence Associated Secretory Phenotype). We have identified several SASP factors that may block immune surveillance by NK cells. The aim of the third project is to develop therapeutic interventions based on removal of these SASP proteins for enhancing immune surveillance of senescent cells in aging and senescence mice model. All these projects involve extensive use of cell culture and mice models and involves working with human blood samples, primary human cells, and mice tissue.

The ideal candidate for this full-time position has a Ph.D. in chemical/biological sciences and substantial bench experience. The candidate must be adaptable, a team player, self-motivated, and interested in problem-solving. The prospective postdoc will work closely with the Primary Investigator (PI) on a couple of projects with the potential of landing senior authorships in research publications resulting from the work. Duties will include performing wet-lab experiments, recording and analyzing data, and presenting the results in meetings every 2-3 months.
